Abstract. Epithelia are cells that have different physical sides with different sets of membrane properties. In this talk, a couple of examples of our work in epithelia will be presented. In the first example, retinal pigment epithelium, a monolayer of cells that underline the photoreceptors in the eye, must properly transport metabolic byproducts (CO2, lactic acid, H2O) from the subretinal space to the choroid. We show that CO2 is significantly more permeable in the apical membrane than the basolateral membrane, and we decipher the intricate interacting CO2 and lactic acid transport mechanisms. Our findings contribute to a better understanding of how the eye functions and help prevent, diagnose, and treat many common eye diseases. In the second example, we present how the kidney epithelia shed proteins into the urine through exosomes and how we have succeeded in assembling a urinary proteomic database. This study demonstrates the potential to diagnose quickly genetic diseases in patients and to profile a large population for personalized medicine.
